So I'm replacing the driver side brake line that runs on the rear axle from the distribution block to the driver side brake. Do I only need to bleed the brake on that side or should I do both rears?

There is always a chance that the fluid left in the good side ran out while you had it disconnected. Bleed both, doing the RR first, then the LR.
