65 Mustang Convert, 289, 3 speed. Going to do mirror tips to get a good tight seal and would like to redo rubber on vent window. Tried to remove when I updated interior years ago. So my question is: do I need to remove main door glass to get the vent window frame out. Last time I wrestled with the thing for hours and could not figure out how to get out, but I had left the main door glass in. So before I try again I am asking for some advice this time. Thanks.

I've found it EZ'er to remove the big glass first.
Like MS told me once..."cover all the edges of the access holes with tape so it won't cut you if/when you need to reach inside the door."
Good time to grease and lube and/or replace the window rollers...WYAIT!
Vacuum iand clean the insides of the doors and lube/adjust the lock mechanism WYAIT.

Remove both of the flanged adjusting bolts as well as the bolts that hold the vent window frame and then slide the vent assembly straight up. It is not required to remove the door glass first.

In addition to what MS said, sometimes it's stuck in there pretty tight and a little judicious leveraging upward from inside the door can help. Also, you might have someone pry apart the outer and inner skins a bit at the top of the door, using a couple of large flat head screw drivers and some small bits of plywood (to distribute the load and protect the paint). When you reinstall it, don't tighten the upper bolts too much as you can break the frame. I think MS has a post on installing door glass you should locate.
