Well, I've got a whopping total of 2 miles on my car now and if anyone remembers from the below thread, I have a problem with wheel fitment on the d/s, although the p/s fits fine. I have attributed the difference in fitment to the difference in factory and re-pop fenders. The d/s rubbed a little bit just coming in and out of my driveway, so I need to do something sooner than later.
My wheels are currently 17x 7 with 4.25" backspace and a 235/45/17 tire. I am going to switch to a 225/45/17 tire and am going to change the backspace on the wheels. So, here is the question... should I go with a 4.5" bs or, to make sure I have no clearance issues, go with a 4.75" bs and then, if I have the room, use a 1/4" spacer?
Thoughts on using spacers?

Avoid spacers since you have the opportunity to do it right, in the first place.
17x7 with 4.5 bs will fit great.

Agree with avoiding using a spacer. Suggest you check the clearance to the as-is inner fenders which you can use as a guide for how much more wheel backspacing you can handle.

I had spacers on my 66 with 2000 Mustang wheels 235/45/17 tires b/s unknown. The car drove like a Buick Electra. I had the fronts narrowed following the specs MS used.

I have extended studs all the way around and run 3/16" spacers (a bit thinner than the ones you are considering). I got the stance I like.
